Damaged DNA generates mutations and genomic instability that ultimately threaten cell or organism viability. Recognition of DNA damage is a very challenging cellular process, which consists in detecting rare modifications to the DNA in a large pool of intact genomic material. The goal of the proposed project is the study of the dynamics of DNA damage localisation, recognition and repair processes in the radiation-resistant bacterium Deinococcus radiodurans. The project will focus on the so-called “nucleotide excision repair pathway”, that is responsible for the removal of a wide variety of structurally
unrelated lesions. The objectives are to investigate the cellular localisation and dynamics of these proteins in vivo using “super-resolution” fluorescence microscopy. In this frame, the so-called
“PALM” super-resolution microscopy technique, based on the detection of single molecules, will be developed so as to carry out quantitative, multi-colour imaging. We expect this work to shed light on the mechanisms used by repair proteins to locate and repair their DNA substrates and, at the same time to contribute to the development of nanoscale fluorescence imaging.

This PhD project is one of the 35 proposed projects offered by the CEA for 2014-2015 as part of the Irtelis PhD programme.
