In adults, the majority of the tumors are from epithelial origin. Carcinogenesis is characterized by the dissolution of the intercellular adherens and tight junctions, followed by the loss of the apico-basal polarity, leading to the transition towards a mesenchymal phenotype and the acquisition of migratory properties. The most aggressive cells will become invasive and form metastases.

The thesis project aims at characterizing the anti-tumoral role of EFA6B, a tight junction regulator, which we have recently identified as an antagonist in breast cancer. The role of EF6AB will be studied using a cell culture model that recapitulates the apparition of cell populations of the "claudin-low" breast cancer subtype from normal human mammary cells. In addition, the tumoral progression of EFA6B genetically modified human mammary cells will be analyzed in vivo by orthotopic xenograft in mice. Besides basic cell culture, biochemistry and molecular biology, techniques used will include 3D-cell culture, confocal imaging, qRT-PCR, etc.
